In the future try using the code tags around your script, makes it easier to read:

IfWinNotActive, Program Manager, FolderView, WinActivate, Program Manager, FolderView
WinWaitActive, Program Manager, FolderView
MouseClick, left, 121, 300

There could be any number of reasons your code doesn't appear to work, but you'd have to explain in more detail what each movement is trying to do in order for us to better understand. It looks like you're taking two steps to get to the Runescape website which may not be necessary. You could try this portion of script based on what I presume you're trying to do:
| Code: | Run http://www.runescape.com{ENTER} ; this skips opening Internet Explorer and clicking into the address bar to enter the website address
WinActivate, RuneScape - the massive online adventure game by Jagex Ltd - Windows Internet Explorer
WinWaitActive, RuneScape - the massive online adventure game by Jagex Ltd - Windows Internet Explorer,
StatusBarWait, Done, , RuneScape - the massive online adventure game by Jagex Ltd - Windows Internet Explorer ;waits until the status bar says 'Done' before executing any further script
... |
Everything below that point you would have to explain what you're trying to do with each set of mouse clicks. It looks like you're just clicking buttons to gain further access into the site, in which case using StatusBarWait to tell you when the page is done loading would be helpful. |
