Reading colorvalue of a partial tranparent screen area within a game

hi, is this possible with AHK in realtime ,inside a game ?

I notice from spywindow that colors of pidel/area changes some according to the background , in a game.

I need to compare colorvalues, using pixelgetcolor or pixelsearch (pref. for a small area, but a pixel can do), for a partly transparent part of gamescreen where game is running in window mode.

Any hints good links for this topic?
